Title : Latest innovations in leprosy diagnosis and therapy: Towards accelerating global elimination

Abstract:

Background: Leprosy (Hansen’s Disease) remains a significant global health challenge, classified as a Neglected Tropical Disease (NTD) with over 170,000 new cases reported in 2024. Although multidrug therapy (MDT) has drastically reduced the disease burden, challenges in early diagnosis, reaction management, and transmission prevention still hinder elimination efforts. This research aims to present a comprehensive review of the latest breakthroughs in leprosy diagnostics and therapeutic modalities that have the potential to transform the disease management paradigm.

Methodology: This review synthesizes data from peer-reviewed research publications, clinical trials, and global health organization reports from the 2024-2026 period. The primary focus is on innovative diagnostic technologies and new therapeutic agents that have shown significant potential in clinical trials.

Results: In diagnostics, significant progress has been made through the development of artificial intelligence (AI)-based tools such as the AI4Leprosy initiative, which uses skin lesion image analysis for rapid and accurate screening. This technology, coupled with advanced molecular tests like multiplex PCR assays and anti-PGL-I antibody detection, promises to dramatically increase diagnostic sensitivity and specificity, even in resource-limited settings. On the therapeutic front, a proof-of-concept study published in the New England Journal of Medicine demonstrated that Bedaquiline monotherapy for 8 weeks successfully achieved M. leprae clearance in multibacillary leprosy patients within 4 weeks. Furthermore, new agents like Dovramilast show potential as a breakthrough treatment for type 2 leprosy reactions, while new prevention strategies such as PEP++ (Post-Exposure Prophylaxis) are being explored to break the chain of transmission.

Conclusion: The landscape of leprosy management is undergoing a transformation driven by innovations in AI-based diagnostic technology and the development of more effective, shorter-duration therapeutic agents. These advancements offer new hope for accelerating the global elimination of leprosy, reducing disability rates, and addressing the associated stigma. This presentation will elaborate on these key findings and discuss their implications for clinical dermatology practice worldwide.

Biography:

Dr. Renni Yuniati, Sp.D.V.E., Subsp.D.T., FINSDV, FAADV, MH is a dermatologist and venereologist affiliated with Dr. Kariadi General Hospital, Semarang, and the Faculty of Medicine, Universitas Diponegoro. She obtained her medical degree from Universitas Sebelas Maret in 1997 and completed her specialization in Dermatology and Venereology at Universitas Diponegoro in 2008. Her academic interests include morbus hansen and dermatologic infections. She also has published several publications internationally, one of them is difference of tumor necrosis factor alpha levels in multibacillary leprosy between reversal reaction and non-reversal reaction patients in Pakistan Journal of Medical and Health Sciences.
